While electron microscopy offers exceptional subcellular detail, it is limited by demanding preparation, restricted field of view, and limited 3D context. Soft X-ray tomography (SXT) overcomes many of these challenges by imaging fully hydrated, cryo-preserved samples in 3D with minimal preparation and native contrast. The lab-scale SXT-100 microscope now enables integration into correlative workflows with light and electron microscopy. We present recent advances in CLEXM workflows, combining cryo-FM, SXT, and FIB/SEM or cryo-ET, highlighting their power for high-resolution, 3D imaging of cells and tissues in near-native state.
